Skopelos heeft ongeveer 4.520 inwoners. (2024) Bij de volkstelling van 2021, had Skopelos-stad 2.940, het dorp Glossa 826 en Neo Klima (Elios) 431 inwoners. In de zomer kan het aantal inwoners door de toestroom van toeristen oplopen tot wel 20.000.
Halverwege de 19e eeuw, bij de eerste volkstelling, was Skopelos-stad na Athene en Nauplion de grootste stad van de nieuwe Griekse staat.
